When you extract a PCR product, its ends are the primers, and it is correct.
But when you do PCR for primers of the extended DNA alphabet, these product ends contain characters of the extended DNA alphabet too. It is wrong and there must be a correct nucleotides instead of ambiguous characters.
Different combination of mismatches and ambiguous characters in primers and sequence must be considered.
